HILIC (Hydrophilic Interaction LIquid Chromatography) is a special case of normal phase chromatography. It is used primarily for separation of very polar compounds. The typical mobile phase for HILIC chromatography includes acetonitrile with a small water content. It is commonly believed that in HILIC, the mobile phase forms a water rich layer on the surface of the polar stationary phase creating a liquid/liquid extraction system. On HILIC mode, polar compounds have longer retention time than non-polar compounds. It avoids using ion-pair reagents or high water content for separation of very polar compounds and provides better peak shapes for basic compounds. The high concentration of organic solvent in the mobile phase pushes up the sensitivity of LC-MSD analysis.

PolyHYDROXYETHYL A - An excellent HILIC
column for biological molecules (From PlolyLC). The column is
excellent for separation of peptides, nucleic acids, carbohydrates, some
proteins. Its great polarity means less organic solvent is needed to get
retention compared to other HILIC columns. In the absence of organic solvent, PolyHYDROXYETHYL A™ functions in
the Size Exclusion Chromatography
(SEC) mode. It comes with different pore sizes (60 - 300 Å) for different
applications.
